Genome editing in cultured fishes

Abstract With external fertilization, high fecundity, and established methods for propagation larval rearing cultured species, fish provide systems well suited to genome-editing procedures. While early experiments utilized zinc-finger nucleases transcription activator-like effector (TALENs), most recent ones have used the CRISPR/Cas9 editor, achieved rates of targeted genomic insertion above those classical transgenic methods, with lower frequencies off-site integration. Genome-editing fishes focused on improving growth rate disease resistance, achievement reproductive confinement, other valued traits. As reviewed here, advances in knowledge key molecular pathways and, some cases, favorable alterations phenotype been achieved. For example, loss-of-function myostatin, a negative regulator muscle growth, led increased mass, greater weight, fillet yield genome-edited lines red sea bream, tiger puffer, Nile tilapia than their unedited counterparts. The bream line become first animal reach commercial production. all animals, wide adoption will depend upon addressing issues regulation, consumer acceptance, breeding infrastructure.
